Functional annotation of non-model bacteria based on sequential homology

Genomová sekvence je důležitým zdrojem informací v oblasti biologie, a proto je neustálým předmětem zájmu vědeckého výzkumu. Na práci s genomem a jeho analýzu nabízí bioinformatika různé výpočetní metody. Tato práce se věnuje bakteriálnímu genomu, jeho organizaci, základním vlastnostem a následně popisuje jeho anotaci. Zaměřuje se hlavně na funkční anotaci (popis biologické funkce predikovaných genů) na základě přiřazení takzvaných klastrů ortologních genů (COG) s využitím sekvenční homologie. Popisuje nejpoužívanější nástroje a databáze, které se využívají pro COG anotaci a poté několik z nich porovnává při anotaci sedmi bakteriálních genomů. Jejím hlavním cílem je navrhnout metodu, která vylepší COG anotaci a zjednoduší její výslednou vizualizaci.
ARP2/3 complex interaction with plant secretory pathway components

The ARP2/3 complex is an evolutionarily conserved protein complex involved in actin nucleation. In animals, the loss of function of ARP2/3 subunits is usually lethal. In contrast, arp2/3 plants are viable and exhibit certain morphological defects. One of the characteristic traits of arp2/3 plants is impaired cell adhesion. This suggests a role for the ARP2/3 complex in plant cell wall secretion, specifically pectins. However, the exact molecular mechanism of ARP2/3 function in this pathway has not been elucidated yet. Preliminary results suggest a possible interaction of ARP2/3 with the secretory pathway proteins GNL1, COPI, and COG. This work combines two methods - TIRFM/VAEM and co-immunoprecipitation - to study the interaction of ARP2/3 with selected markers. However, neither colocalization nor physical interaction was demonstrated. This work also characterises COPI, COG and GNL1 with TIRFM/VAEM and shows that there is an interaction between GNL1 and endocytic markers. Key words: ARP2/3, COPI, COG, GNL1, secretory pathway
Hip joint stability and injury prevention in sport

The aim of this thesis is to describe, how does the strengthening of stabilization hip muscles affect whole postural stability and at the same time how does it affect standing on one leg for athletes in different sport. The theoretical part of the thesis summarizes information about anatomy a kinesiology of hip joint. Includes important angles measured in the age of hip joint development. Besides is described postural stability, and stability function in the various levels on human body with all the participated muscles while hip joint stabilization. In the practical part is research of three probands doing cross country skiing, athletics, and triathlon. Measurement was consisting of posturography, kinesiological analysis and by performing special tests focused on hip joint. All the patients received paper of exercises to do at home with addiction of individual exercises for patient's pathology. At the end of the therapy was compared all the information from the first and second measurement and done conclusion. The resulting effect was positive, because in most patients was improvement of secondary performing test measuring stability, placement of COG and lateral stability of hip joint. Another result was subjective evaluation in the way of dialogue. Patients evaluated the therapy as a positive for them and wanted to continue in cooperation.

Evaluation of postural stability of athletes - a literature review

Title: Evaluation of postural stability of athletes - a literature review Objectives: Thesis will address retrieval processing rating Dynamic Postural Stability of athletes . Compares analysis or results of research or investigations of different laborers working with this theme. It also mentions tests and machines used in clinical practice. It also investigates in what sports was postural stability tested and how or with what results. The most advanced, currently the most recognized dynamic posturography with impartial value is EquiTest device from NeuroCom. Therefore, the practical part focuses primarily on the results of examination of this device and make statistics about that. Methods: The diploma thesis has descriptive and analytical character. It is elaborate as a literary review . Results: The study includes 142 studies, of which only 11 concern the assessment of postural stability of athletes. Sports disciplines include tennis, football, baseball, tai-chi, taekwondo, gymnastics, basketball, softball.
